4% PFA (Paraformaldehyde) - ml  


4% paraformaldehyde in 1 X phosphate buffered saline - pH 7.4


Reagents needed:

10
g paraformaldehyde
50
μl 10N NaOH
25
ml ddH2O

** CAUTION ** Paraformaldehyde Fumes Are Toxic. All Work Should Be Done In A Ventilated Fume Hood.

Directions:

1) Heat 200 ml of ddH2O to 60°C (either on a hot plate or in a microwave oven).

2) Add 10 g of paraformaldehyde while stirring the 60°C ddH2O.

3) Add 50 μl of 10N sodium hydroxide (NaOH), continue to stir.

4) Allow solution to stir until paraformaldehyde dissolves.

5) Add 25 ml of 10X PBS and mix.

6) Add ddH2O to a final volume of 250 ml.

7) Filter 4% PFA through a 0.45 μm filter.

8) Aliquot and freeze at -20 °C for long-term storage.
